Is the caller legit, if they didnt know the Jewish Holiday is over?
Does Chai Lifeline use non-Jews for these phone calls?

I know several secular Jews who have volunteered for Chai Lifeline. I can imagine that if they have been in the habit of calling with this greeting for days in a row, it might just naturally slip out for a few days more - maybe even for a frum person, but especially for someone who might not be lighting and counting out each night.

Sorry to disillusion you but Chai lifeline is a business. They have all manner of people volu terring. While they do lots of good for lots of people, there are agendas and politics, like all big businesses.
This, from a family who needed their help.
So, to answer your question, it's most likely legit.
