Its for my sister with 6 kids- 9th grade through 3 yr old..budget not such an issue since its only one day there.

Museum Campus -- museum of science and industry or the Shedd Aquarium, maybe with that range of ages I'd prefer the She'd Aquarium for the 3 year old. Be sure to get tickets to the show and any special attractions when you come in as they fill up.

The Museum of Science and Industry and Shedd Aquarium are both excellent, and would definitely fill up an entire day. If she can afford it, I would go early in the morning to the Sears/Willis Tower and go up to the top - before the line gets long, as that's a truly iconic Chicago experience. I would then explore the area around it on Michigan Avenue along with the Bean and Crown Fountain, and then take an Uber to either the science museum or the aquarium.
